摘要
目的:比较核酸富集处理前后3种核酸提取方法对精炼食用植物油的提取效果。方法:基于核酸具有水溶性的原理,将大量植物油样中的核酸最大可能的萃取富集到少量具有细胞裂解功能的水相中,再以此水溶性液体为对象,采用两种试剂盒法和经典的CTAB核酸抽提方法进行后续提取步骤,以植物叶绿体tRNA基因片段的扩增结果作为各种方法核酸提取效果的评估指标。结果:核酸富集处理前,采用3种核酸提取方法在其核酸提取液中都未扩增出叶绿体tR-NA基因片段。核酸富集处理后,在12个精炼植物油中,采用DNeasy Plant Mini Kit试剂盒法,只有3个样品的核酸提取液未扩增出目的片段;采用Food EX Magnetic试剂盒法,有6个样品的核酸提取液扩增出目的片段;采用CTAB法,只有2个样品的核酸提取液未扩增出目的片段。结论:采用核酸富集处理后,3种提取方法的核酸提取效率都显著提高。
Objective:To validate the difference in DNA extraction efficiency of 3 DNA isolation methods for refined vegetable oil samples before and after using nucleic acid enrichment treatment.Methods:Based on the principle of nucleic acids with water-solubility,the nucleic acids in large amounts of vegetable oil samples were enriched in small amounts of aqueous phase by abstraction.Then the aqueous phase was used to isolate DNA according to 3 extraction methods.The assessment of nucleic acid extraction results was based on the amplification results of plant chloroplast tRNA gene fragment.Results:Before nucleic acid enrichment treatment,using any of the three kinds of methods,no chloroplast tRNA gene fragment was amplified.After nucleic acid enrichment treatment,for 12 refined vegetable oil samples,the target fragment was not amplified in only 3 samples by DNeasyPlant Mini Kit method;the target fragment was amplified in 6 samples by Food EX Magnetic Kit method;and the target fragment was not detected by PCR in only two samples using CTAB method.Conclusion:After nucleic acid enrichment treatment,nucleic acid extraction efficiency has been improved significantly whatever extraction methods were used.
