固相萃取-高效液相色谱法测定水产品中的三聚氰胺

Determination of melamine in aquatic products by SPE-HPLC

摘要 目的:建立用固相萃取-高效液相色谱法测定水产品中三聚氰胺的高效液相色谱法。方法:水产品中的三聚氰胺经阳离子交换固相萃取柱净化后,采用Kromasil C18色谱柱(4.6 mm×250 mm,5μm)分离,以庚烷磺酸钠-柠檬酸缓冲液:乙腈(85:15)为流动相,由高效液相色谱-紫外检测器分析测定。结果:该方法在10~100μg/ml的范围内具有良好的线性关系(r=0.9993);加标回收率为80.1%~88.9%;RSD为2.1%~5.0%;方法检出限为1.6 mg/kg。结论:该方法操作简便快捷、净化富集效果好、重现性较高,能满足实际样品的日常检测需求。 Objective:To establish a SPE-HPLC method for the determination of melamine in aquatic products.Methods:Melamine in aquatic products was cleaned-up by SPE column,then separated by Kromasil C18 column(4.6 mm×250 mm,5 μm)with ultraviolet detection.The mobile phase consisted of sodium heptanesulfonate-citric acid buffer solution:acetonitrile(85:15).Results:The linear ranges were 10~100 μg/ml(r=0.9993),the recoveries were in the range of 80.1%~88.9%,the relative standard deviations were 2.1%~6.1%,and the determination limit was 1.6 mg/kg.Conclusion:This method is of good reproducibility and simple operation and it could be applied to the determination of the aquatic products.
