摘要
目的研究Cdc42基因在新疆维吾尔族食管癌组织中的表达并探讨其在维吾尔族食管癌发生、发展中的作用。方法采用逆转录多聚合酶链反应技术检测Cdc42基因在20例食管癌组织及其相应癌旁组织中的表达情况,并与临床病理资料结合分析其意义。结果癌组织与相应癌旁组织相比,在20例癌组织中,有15例表达阳性,占75%;癌组织表达量明显高于癌旁组织的有7例,占35%;其中有淋巴结转移的癌组织中Cdc42 mRNA的表达率为80%(8/10),无淋巴结转移的为60%(6/10);肿瘤浸润至外膜层的表达率为80%(8/10)。结论提示Cdc42基因在转录水平有的异常表达,Cdc42基因在不同分期的癌组织中,Cdc42基因高表达与淋巴结转移和肿瘤浸润深度有关。Cdc42基因在癌组织和不同分期癌组织中表达的改变,提示它所参与的细胞凋亡及细胞周期过程的变化在维吾尔族食管癌的发生、发展、转移、浸润过程中起了一定的作用。
Objective To study expression of apoptosis - related gene Cdc42 in Uygur esophageal carcinoma tissue and discuss its significance in genesis and development of Uygur's esophageal cancer. Methods The expression of Cdc42 gene mRNA was detected in 20 cases with esophageal cancer tissues and tumor - adjacent normal tissues by Reverse Transcription Polymerase Chain Reaction, and the significance was analyzed by combining with clinic - pathological variables. Results The overall positive rate of Cdc42 gene mRNA expression in the 20 cases of esophageal cancer tissues was 75%. The frequency of hMLH1 expression was 80% with lymphoid node metastasis, 60% in those without metastasis and 80% in T3 stage carcinoma. Conclusions It is suggested that the abnormal expression of Cdc42 gene affect the occurrence of Uygnr's esophageal cancer mainly in transcriptional level, Cdc42 gene play important role in carcinogenesis and development of Uygur's esophageal cancer.
