摘要
目的:研究清胰化积中药对人胰腺癌SW1990体内生长的作用,并探索其对代谢相关基因表达的调节作用。方法:荷SW1990胰腺癌裸小鼠随机分为对照组、吉西他滨化疗组(化疗组)、中药高、中、低剂量组,实验结束后测量瘤质量。提取对照组及中药组肿瘤组织mRNA,制备cDNA探针,与基因表达谱芯片进行杂交,得出药物作用后表达有差异的代谢相关基因。结果:清胰化积中药高、中剂量组及化疗组瘤质量低于对照组,具有显著性差异(P<0.05)。筛选出清胰化积中药作用后表达上调的代谢相关基因3α-羟基类固醇脱氢酶基因,表达下调者包括脑糖原磷酸化酶、对氧磷酯酶、中性α-葡萄糖苷酶基因。结论:清胰化积中药对人胰腺癌SW1990具有一定的抑瘤作用,可能与其对代谢相关基因的调节有关。
Objective:To study the effect ofthe herbal decoction QingyiHuaji Formula(QYHJ) on the proliferation ofhuman pancreatic cancer cell line SW1990 in vivo and to explore the regulation effect ofthe decoction on metabolism related gene expression.Methods:Tumor burdened nude mice were randomized into control group,gemcitabine group, and QYHJ groups at different dose.After treatment, tumor weight was calculated.Tumor mRNA ofthe control group and the QYHJ group was extracted.The fluorescent cDNA probes were prepared, hybridized with cDNA microarray.The metabolism related genes with different expression were screened through the analysis ofgene expression profile.Results:Tumor weight ofthe QYHJ groups at high and middle dose was lower than that ofthe control group(P0.05).4 metabolism related genes were identified, including the up-regulated gene 3-αhydroxysteroid dehydrogenase, and the down-regulated genes brain glycogen phosphorylase, paraoxonase-2, and neutral α-glucosidase.Conclusion:QYHJ decoction may inhibit the proliferation ofpancreatic cancer SW1990 in vivo, and may partly by modulating the expression ofmetabolism related genes ofcancer cells.
