摘要
目的:从海马区超微结构及抗氧化损伤等方面探讨低浓度铅接触行为记忆改变及中药驱铅丸的干预机制。方法:32只大鼠随机分为4组:空白组饮双蒸水;其他3组饮水中均添加0.02%醋酸铅,其中中药组按驱铅丸3.5g·kg-1·d-1的剂量2ml/d灌服;Ca-EDTA治疗组(依地酸钠钙加普鲁卡因肌肉注射,50mg·kg-1·d-1,连续注射4d为1个疗程,休息4d后进行下一个疗程,连续治疗7个疗程);病理组不予任何治疗。60d后,进行Morris水迷宫试验,检测全血、脑组织铅含量;检测脑组织MDA、SOD和GSH-Px含量;用电镜检测海马组织超微结构。结果:与空白组比较,病理组血铅、脑铅含量显著增高(P<0.01),与病理组比较中药组脑铅、血铅含量显著降低(P<0.05);Morris水迷宫试验结果提示病理组大鼠空间定位航行能力下降(P<0.05),中药组定位能力较病理组明显提高,具有统计学意义(P<0.001);病理组MDA显著高于空白组(P<0.01);与病理组比较,各治疗组MDA含量均显著降低(P<0.05),与病理组比较,中药组SOD、GSH-Px含量显著增高(P<0.05)。结论:驱铅丸可以改善铅接触大鼠的行为记忆障碍,其机制与驱铅丸改善铅导致的氧化损伤有关。
Objective:To investigate the effect oflow-dose lead on oxidative damage and changes ofhippocampus ultrastructure and the interference ofQuqian Pills on it.Methods:32 male Wistar rats were chosen and randomly divided into 4 groups, 8 rats respectively:the control group was given distilled water orally,the model group and other two groups were given 0.2% Lead Acetate orally.In addition, the Quqian Pills group was treated by Quqian Pills 3.5g?kg-1?d-1, 2ml/d, the Ca-EDTA group was treated by intramuscular injection ofcalcium disodium ethylenediaminetetracetic acid, 50mg?kg-1?d-1, 4 days in a row injection for a course oftreatment, and taking a rest for 4 days, then into another course oftreatment, 7 courses oftreatment.After 60 days, the Morris test was used to measure the ability oflearning and memory, the graphite heat atomic absorption spectrum method was used to determine the lead concentration in blood and brain, the concentration ofMDA, SOD andGSH-Px.Results:①The ability ofdimensional memory in pathologic group declined significantly compared with control group and the Quqian Pills group(P〈0.05) by Morris test.②The lead concentration in blood and brain in lead poisoned model group increased significantly compared with others(P〈0.05).③Compared with the control group, the concentration ofGSH-Px and SOD in the lead exposed rats decreased significantly(P〈0.05), MDA in hippocampus increased(P〈0.05), and SOD, GSH-Px ofthe Quqian Pills group rats increased and MDA decreased significantly(P〈0.05) compared with the pathologic group.Conclusion:The Quqian Pills could reduce the behavior and memory obstacle induced by lead.It may be correlated with that the Quqian Pills could improve the oxidative damage caused by lead.
