黄芪多糖对大鼠慢性阻塞性肺疾病羟脯氨酸和基质金属蛋白酶-9的影响 被引量:6

Influence of astragalus polysaccharide on hydroxyproline and MMP-9 in rats with COPD

摘要 目的观察黄芪多糖对慢性阻塞性肺疾病(COPD)大鼠肺组织中羟脯氨酸和基质金属蛋白酶-9(MMP-9)的影响。方法采用香烟熏吸加气管内注入脂多糖(LPS)法建立大鼠COPD模型。黄芪多糖高、中、低3个浓度及强的松灌胃干预30 d。生化法检测肺组织中羟脯氨酸含量,RT-PCR法检测MMP-9基因表达水平,免疫组化法半定量检测MMP-9蛋白含量。结果模型组大鼠肺组织羟脯氨酸含量和MMP-9表达水平较正常对照组明显增加(P<0.01);黄芪多糖干预后,羟脯氨酸呈剂量依赖性明显减少(P<0.05,P<0.01),MMP-9基因及蛋白表达水平亦明显降低,以中剂量效果最明显。结论黄芪多糖可减少COPD大鼠肺组织羟脯氨酸的含量和MMP-9的表达,这种作用与改善COPD的肺损伤程度有一定关系。 Objective To observe the influences of astragalus polysaccharide(APS) on hydroxyproline and matrix metalloproteinase-9(MMP-9) in rats with chronic obstructive pulmonary disease(COPD). Methods The rat model of COPD was established by using the method of cigarette smoking combined with intratracheal injection of lipopolysaccharide(LPS).APS(in dose of high,middle and low) and prednisone were given to the rats intragastrically for intervention for 30 days.The lung content of hydroxyproline was detected by using biochemistry method,the gene expression of MMP-9 was detected by applying reverse transcription polymerase chain reaction(RT-PCR),and the protein content of MMP- 9 was determined semi-quantitatively by using immunohistochemistry method.Results In the model group the content of hydroxyproline and MMP-9 expression increased significantly compared with those in the normal control group(P〈0.01).After the intervention of APS the content of hydroxyproline decreased significantly in a manner of dose-dependent,and the gene expression and protein level of MMP-9 decreased significantly,especially when APS was in middle dose.Conclusion APS can decrease the lung content of hydroxyproline and MMP-9 expression in rats with COPD,which may be related to that APS can relieve the lung lesion.
