帕金森病模型大鼠纹状体mGluR5的表达变化及意义 被引量:2

摘要 目的:探讨PD模型大鼠纹状体mGluR5的表达变化及意义。方法:将SD大鼠随机分为正常对照组,PD模型组和亚型选择型mGluR5拮抗剂SIB-1893+PD组,通过免疫组织化学方法观察多克隆抗体mGluR5在大鼠纹状体的表达变化。结果:正常对照组中大鼠纹状体有丰富的mGluR5表达;PD模型组中大鼠纹状体mGluR5表达明显下降;在亚型选择性mGluR5拮抗剂SIB-1893+PD组中,大鼠纹状体mGluR5表达又上调。结论:PD模型大鼠纹状体mGluR5表达下降可能是中枢神经系统在DA能神经缺失以后引起的一种病理性代偿反应,mGluR5可能参与了PD的发生、发展过程。亚型选择性mGluR5拮抗剂有助于减低间接通路的传导,对DA神经元有保护作用,可望成为治疗PD的一种副作用小的新药。 Objective: To explore the expression of metabotropic glutamate reeeptoe subtype 5 (mGluR5) in the striatum of normal and Parkinson's disease model rat and the potential mechanism of the functional disturbance of striatum recognition . Methods :All rats were divided randomly into three groups: normal group, PD group , and SIB-1893 ( antagonist of mGluR5) + PD group. Polyclonal antibody specific of mGluR5 was used to observe the immunoreaetion of mGluR5 in the striatum. Immunobistoehemistry combined with the image quantitative analysis was used. Results :The expression of mGluR5 in the rats of A group was high in the striatum. Whereas an initial down regulation of mGluRS was observed in all the regions of the striatum of rats from B group. At the same time the expression of mGluR5 in the group of C. was increased compared to that in group B. The expression of mGluR5 is mainly on membrane. Conclusions: The expression of mGluR5 in the rats of PD model in striatum was descend. Probably a kind of endogenous protection system maybe take place in the central nervous system after the absence of DA-contained neurone. The expression of mGluR5 is up-regulated through the use of antagonist of mGluR5 SIB 1893 indicated the protective effect on neurone. It can reduce the indirect pathway and will be a new method for clinical treatment.
