摘要
目的:探讨瑞舒伐他汀干预对压力超负荷所引起大鼠心肌肥厚及结缔组织生长因子(CTGF)表达的影响。方法:SD大鼠随机分为5组,各组(除假手术组外)通过部分结扎大鼠腹主动脉构建心肌肥厚模型,A为假手术组,B为单纯手术组,C为卡托普利组,术前一周给予卡托普利(50mg/kg.d)灌胃直至术后4周,D、E组为瑞舒伐他汀组,术前一周分别给予瑞舒伐他汀(2mg/kg.d组及4mg/kg.d组)灌胃直至术后4周。同时A、B组给予等量生理盐水灌胃。测动脉血压,心脏质量指数(HMI),应用病理学方法检测胶原容积百分比(CVF)。用免疫组化法检测心肌组织CTGF蛋白表达,用RT-PCR法检测心肌组织中CTGFmRNA水平的表达。结果:1B组较A组动脉血压、HMI明显增加(P<0.01)、CVF明显增加(P<0.01);D、E组较B组心脏质量指数明显降低(P<0.01),胶原容积百分比降低(P<0.01);C组较A组其HMI、CVF明显降低(P<0.01)。2B组CTGF蛋白及CTGFmRNA表达较A组增加(P<0.01);D、E组CTGF蛋白及CTGFmRNA表达较B组明显下降(P<0.01);C组较B组其CTGF蛋白、CTGFmRNA表达明显下降(P<0.01)。结论:1心肌肥厚大鼠心肌组织中CTGF表达显著增加。2瑞舒伐他汀能够有效地延缓压力负荷诱导的心肌重构并降低CTGF蛋白、CTGFmRNA表达。
Objectives :To observe the effects of rosuvastatin on the hypertrophic myocardium and CTGF of pressure-overloaded rats Methods :SD-rats were divided into 5 groups randomly. Group A. for sham group was given ig normal salin. B. for control group was given ig normal salin C. for captorpil group was given ig. captorpil 50 mg/kg · d for 5 weeks. D. for rosuvastatin low dose group was given ig rosuvastatin 2 mg/kg · d for 5 weeks. E. for rosuvastatin high dose group was given ig. rosuvastatin 4 mg/kg · d for 5 weeks . Rat model of cardiac hypertrophy was established by incomplete constriction of abdominal aorta. Blood pressure and Heart mass index were measured ,pathological method was taken to measure the CVF. Immunohistology for CTGF of the myocardium. RT-PCR was taken to detect CTGFmRNA. Results.. 1. An obvious increase of the Blood pressure and HMI(P〈0. 01) in the control group were detected compaired with the sham group. The rosuvastatin group and the captorpil group showed decrease compared to the was control group in Blood pressure and HMI and CVF(P〈0. 01 ), 2. The control group were increased to the sham group in the expression of CTGF protein, CTGFmRNA, (P〈0. 01) ;An obvious decrease of the rosuvastatin group to the control group in CTGF protein and CTGFmRNA (P〈0. 01). The captorpil group decreased in th expression of CTGF, CTGFmRNA to that of the control group. Conclusion :(1)There is obvious increase in the expresion of CTGF in hypertropic myocardiun. (2) Rosuvastatin can effectively ammeliorate the myocardium hypertropy, and decrease the expression of CTGF, CTGFmRNA.
