分光光度法测定西红柿中总黄酮含量的方法比较 被引量:14

摘要 通过对振荡和超声波法提取以及氯化铝法、硝酸铝显色法测定西红柿中总黄酮的方法进行比较,确定以超声辅助提取、氯化铝显色法测定西红柿中总黄酮含量。结果表明,以芦丁为标准品,在0.00125~0.0020mg/ml范围内线性良好,R2为1,平均加标回收率为95.3%,在最佳提取条件下,实验稳定性、精密度和重现性参数均良好。经实际样品测定证明,此方法测定西红柿中总黄酮效果较好。 In order to establish a reliable method for determining total flavoniods from tomato,oscillation or sonication for extracting total flavonoids and colorimetric method using aluminum chloride or aluminum nitrate for determining the content of flavonoids were compared.Results indicated that sonication extraction and aluminum chloride method revealed an excellent effect on the determination of total flavonoids from tomato.When rutin was chosen as the standard sample,a good linear relationship was observed in the range of 0.00125-0.0020 mg/ml with a correlation coefficient of 1.The average recovery rate of spiked samples using this method was 95.3%.Moreover,stability,precision and reproducibility of this method were also good.The average content of total flavonoids in tomato was 0.44 mg/g through 75% alcohol and ultrasonic treatment.This method will be widely applied to determine total flavonoids due to its simple operation,excellent accuracy and high reproducibility.
