摘要
通过纤维蛋白平板初筛、纤维蛋白原平板和摇瓶复筛以及传代培养,从采自广西北海的海泥中筛选出一株具有稳定遗传性能的高纤溶酶酶活力菌株HQS-3。对该菌株进行形态、生理、生化鉴定及16SrDNA序列分析的结果表明该菌株为枯草芽孢杆菌,该菌株产纤溶酶初始酶活高达1980U/mL,传代10次后酶活保持稳定。经溶血性实验对纤溶酶的安全性进行研究,结果表明,纯化后的纤溶酶并未发现有溶血现象。
A strain HQS-3 with high fibrinolytic enzyme activity was,which isolated from the sea mud which derived from Beihai of Guangxi,by using fibrin-plate method as primary screening assay,fibrinogen-plate method and conical flask fermentation as second screening assay. All results fom morphological,physiological and biochemical charaterization,the 16S rDNA sequence analysis identified that the strain was Bacillus subtilis. The strain showed a high initial fibrinolysis activities which was about 1980 U/mL,and it had genetic stablity on plasmin production by lO-generation cultivation. The test of haemolysis was performed for determining the security of the fibrinolytic enzyme, and the result suggested that purified enzyme cannot destroy blood cell.
