残胃癌组织差异蛋白表达谱的初步研究

Proteome Study on the Differences Protein Expression in the Remnant Gastric Cancer Tissues

运用蛋白质组学方法建立残胃癌和癌旁正常胃黏膜组织(pH 3～pH 10)蛋白质表达谱,分析残胃癌和癌旁正常胃黏膜组织中蛋白质表达的差异。通过双向凝胶电泳(2-DE)分离残胃癌组织和癌旁正常胃黏膜组织的总蛋白,并采用凝胶染色和Powerlook 2100XL扫描仪扫描凝胶以获得残胃癌和癌旁正常胃黏膜组织蛋白质表达谱,最终将蛋白质表达谱图经ImageMaster2D Platinum软件加以分析,得到差异表达蛋白结果。通过比较分析3对残胃癌和癌旁正常胃黏膜组织(pH3～10)蛋白质表达谱,共发现108个差异表达的蛋白质斑点,显著差异的蛋白质斑点38个,其中仅在残胃癌组织中表达的蛋白质点3个,残胃癌组织中失表达的蛋白质点7个,15个点在残胃癌组织中表达上调,13个点在残胃癌组织中表达下调。残胃癌与癌旁胃黏膜组织之间存在着显著的蛋白质表达差异。利用蛋白质组技术寻找残胃癌与癌旁正常胃黏膜组织的差异表达蛋白,将为阐明残胃癌发病的分子机制及发现残胃癌的诊断蛋白奠定了基础。

To establish the two-dimensional gel electrophoresis pH 3 - pH 10 maps of human remnant gastric carcinoma and paired paracancerous mucosa, and to analyze differently expressed proteins of human remnant gastric carcinoma and paired paracancerous mucosa, the total proteins of human remnant gastric carcinoma and paired paracancerous mucosa were extracted, separated and analyzed by two dimensional gel electrophoresis （2 - DE）. Two-dimensional gel electrophoresis （2 - DE） was followed by ImageMaster 2D Platinum software analysis and used to screen changes in protein patterns of extract. By comparing and analyzing pH 3 - pH 10 protein expression maps of 3 cases of remnant gastric carcinoma and paired paracancerous mucosa, 108 differently expressed protein spots were detected. There were 38 protein spots that displayed quantitative changes in expression in the maps, of which 3 expressed only in remnant gastric carcinoma, 7 in normal gastric mucosa tissues, 15 higher and 13 lower in remnant gastric carcinoma than in paracancerous mucosa tissues. Proteins express differently in remnant gastric carcinoma from paired paracancerous mucosa. Searching for differently expressed proteins between the two types of tissues may throw light on the molecular mechanism of remnant gastric carcinoma.
