氯化高铁血红素对正常及再生障碍性贫血小鼠红系祖细胞的体内诱导作用 被引量:3

In vivo inductive effect of hemin on the erythroid progenitor of the normal and aplastic anemia mice

摘要 目的:研究氯化高铁血红素(Hm)对正常及再生障碍性贫血(再障)小鼠红系祖细胞(BFUE和CFUE)的体内诱导作用。方法:分不同剂量诱导组(0.65,6.50,13.00μg/g体重,诱导时间为静脉注射后6小时)及不同时间诱导组(注射后6,12及24小时,Hm剂量为6.50μg/g体重),采用甲基纤维素体系进行红系祖细胞培养。结果:再障小鼠骨髓BFUE数量(3±2个/股骨)及CFUE数量(12±3个/股骨)与正常小鼠相比均明显减少[BFUE为1056±160个/股骨,P<0.01;CFUE为12±3个/5个高倍视野(×200)]。6.50μg/g体重Hm诱导后6小时,再障小鼠BFUE数量增加到10±2个/股骨,CFUE增加到39±12个/股骨,与对照组再障小鼠相比,均有显著差异(P<0.01)。经诱导后的正常小鼠骨髓BFUE增加到3410±708个/股骨,CFUE数量增加到38±3个/5个高倍视野,与对照组正常小鼠相比,有显著差异(P<0.01)。Hm体内诱导的最适剂量为6.50μg/g体重,最适诱导时间为静脉注射后6小时。结论:Hm是一种体内红系细胞分化诱导剂,对再障小鼠红系祖细胞亦有诱? Objective:To investigate the in vivo effect of hemin on erythroid progenitor of the normal and aplastic anemia mice. Methods:BFU E and CFU E of normal and aplastic anemia(AA) mice were assayed at different time (6h, 12h and 24h) after hemin injection on a dosage of 6.50μg/g body weight, and at 6h after injection of different doses of hemin (0.65μg/g, 6.50μg/g and 13.00μg/g body weight). Results: The yields of BFU E and CFU E from AA mice were reduced compared with that from normal mice. At 6h after hemin injection (6 50μg/g b.w.), the yields of BFU E and CFU E from AA mice were increased from 3±2/femur and 12±3/femur to 10±2/femur (P<0.01) and 39±12/femur (P<0.01), respectively, and so did the case for normal mice (for BFU E, from 1056±160/femur to 3410±708/femur,P<0.01, and for CFU E, from 12±3/five 200×fields to 38±3/five 200× fields, P<0.01). Low dose of hemin had no inductive effect. Conclusion: Hemin is a in vivo inducer of erythroid progenitors both in normal and AA mice.
