摘要 目的:研究辅酶Q10对紫外线致角膜上皮细胞光损伤的防护作用及其机制。方法:Wistar大鼠48只随机分为正常对照组、阳性对照组与辅酶Q10治疗组。紫外线照射建立角膜上皮紫外线光损伤模型。治疗组于光照前和光照期间每天辅酶Q10灌胃,共21d。阳性对照组每天给予等体积的生理盐水灌胃,正常对照组不给任何处理。紫外线照射第3次后12h处死动物取角膜,光学显微镜下观察各组角膜组织病理学改变,分光光度法测量角膜上皮组织的SOD,MDA的含量,并进行统计学检验。结果:角膜上皮紫外线光损伤后阳性对照组SOD水平下降,MDA水平升高,其差异与正常组比较有统计学意义(P<0.01)。与阳性组比较,治疗组SOD水平升高,MDA含量降低(P<0.05)。结论:辅酶Q10对角膜上皮紫外线光损伤具有保护作用,机制可能与清除自由基和增强抗氧化能力有关。 AIM: To explore the protective effect of coenzyme Q10 on ultraviolet (UV) irradiation-induced injury of rats corneal epithelium and its mechanisms.METHODS: Forty-eight Wistar rats were divided randomly into normal control group, positive control group and coenzyme Q10 group. The UV irradiation-induced damage model of Wistar rats was established by exposing to ultraviolet. The rats in coenzyme Q10 group were intragastric administrated by coenzyme Q10 every-day for three weeks before and during UV exposure. The rats in normal group did not received ultraviolet light, and the rats from positive group only underwent ultraviolet ray without the administration of drug. Histopathologic changes were observed with light microscopy. The contents of malondialdehyde (MDA), superoxide dismutase (SOD) in the corneal epithelium were measured by photometry, Statistical analysis was used to assess results.RESULTS: After UV irradiation-induced injury, the content of MDA in positive control group was increased and activity of SOD was decresed obviously, which significantly differed from the normal control group (P〈0.01). In contrast to the positive group, the content of MDA in coenzyme Q10 group was reduced and activity of SOD was increased (P〈0.05).CONCLUSION: Coenzyme Q10, by removing oxygen free radicals, elevating the ability of anti-oxidation in corneal epithelium, can protect corneal epithelium against UV irradiation-induced injury.
