缺血性视神经病变视网膜神经节细胞凋亡过程中c-fos基因改变 被引量:2

摘要 目的:研究缺血性视神经病变以抑制视神经节细胞损伤。方法:Wister大鼠80只,分为正常组和缺血性视神经病变组。缺血性视神经病变组按缺血时间不同分为0.25,0.5,1,2,4,6,12h等7个组。采用RT-PCR方法检测缺血诱导的大鼠视网膜内不同时间c-fos基因的表达情况。结果:正常大鼠视网膜c-fosmRNA有少量表达,缺血可诱导c-fosmRNA的表达,缺血0.25h后c-fosmRNA的表达水平即开始增加,在1h左右达到最高,然后逐渐减少,12h后恢复到正常水平。结论:急性视网膜缺血,c-fos基因表达为快速一过性。 AIM: To prevent retinal ganglion cells (RGCs) from damage by studying the ischemic optic neuropathy (ION)models to conquer the blinding ophthalmocace.METHODS: Eighty Wister rats were divided into 2 groups: the control and the ION group. The ION group included 7 subgroups according to time 0.25, 0.5, 1 hour, 2, 4, 6 and 12 hours. RT-PCR method was used to determine the c-fos gene expression of ischemic-induced RGCs at different times.RESULTS: Small amount of c-fos gene was detected in the control through RT-PCR method and the ischemia can increase the expression of c-fos mRNA at 0.25 hour and peaks at 1 hour and then gradually decreased to normal at 12 hours.CONCLUSION: The c-fos expression in the acute ION models is a fast and transient progress.
