烫伤后创面脓毒症大鼠肝脏PPARα mRNA的表达

摘要 目的探讨烫伤后创面脓毒症大鼠肝脏过氧化物酶体增殖物激活受体αmRNA的表达规律及意义。方法60只背部30%Ⅲ度烫伤大鼠随机分为创面脓毒症组和烫伤对照组,创面脓毒症组大鼠烫伤后10min创面涂布铜绿假单胞菌建立创面脓毒症模型,通过荧光定量PCR法测定伤前、伤后24h、48h、96h及7d的PPARαmRNA表达变化。结果大鼠烫伤后肝组织PPARαmRNA的表达在伤后早期轻度增强,此后表达明显下调,创面脓毒症组下降趋势更明显,致伤后96h表达量降低至最低点,并显著低于对照组(P<0.05)。结论烫伤后创面脓毒症的发生引起肝脏PPARαmRNA表达下调,这种变化可影响肝脏脂质的代谢过程。 Objective To investigate the expression of peroxisome proliferator activated receptor α (PPAR α) mRNA in liver of scalded rats with wound sepsis. Methods Sixty male Wistar scalded rats with 30% total body surface area (TBSA) full-thickness burn were randomly divided into wound sepsis group and scald group (control group). Pseudomonas aeruginosa (10^9/mL) was inoculated to the wounds of rats in sepsis group. The life conditions, appearance of wound, quantity of bacteria in subeschar tissue and pathological changes were observed, whih conformed that the rat model of burn wound sepsis was successfully established. Fresh liver tissues were obtained. Total RNA was isolated from liver tissues with Trizol. PPAR α level in liver was measured by reverse transcription polymerase chain reaction (RT-PCR). Results The PPAR α mRNA expression level was significantly lower in the sepsis group than in the control group 24-96h after scalding(P〈0.05). Conclusion Burn wound sepsis can decrease the expression level of PPAR α mRNA in liver, which might be involved in the initiation and development of lipid metabolism disturbance.
