摘要
木聚糖酶是可将木聚糖降解为低聚糖和木糖的一类酶的总称。它广泛地存在于各种微生物中,在饲料、造纸、食品医药以及能源工业中有着广阔的应用前景。用粗木聚糖为唯一碳源从土壤样品中分离得到了放线菌菌株Xylan009,对其进行了木聚糖酶活性测定,发现菌株Xylan009木聚糖酶活性较好。16S rRNA基因分析显示,菌株Xylan009和拟无枝酸菌属(Amycolatopsis)的菌株的进化距离最近。细胞化学分类数据支持把菌株Xylan009归入到拟无枝酸菌属中。综合菌株Xylan009的形态学特征、生理生化特性、细胞化学分类特征和基于16S rRNA基因序列的系统发育分析结果,确定菌株Xylan009为拟无枝酸菌属的一个新菌种。
Xylanase can degrade xylan into a series of oligosaccharides and xylose. Such enzymes can be produced by many kinds of microorganisms and illustrate a good prospect of application and extension in prepared animal feeds manufacturing, paper-making industry ,food products, medicine and energy industries.The strain Xylan009 was isolated from a soil sample using the selective isolation medium containing the crude xylan as the carbon source.Determination of enzyme activity demonstrated that the strain Xylan009 showed strong xylanase activity.iriS rRNA gene sequences comparison revealed that the most related evolutionary neighbors of this isolate in GenBank were the members of the genus Amycolatopsis.Chemotaxonomic data of the strain Xylan009 supported to affiliate it to the genus Amycolatopsis.Combined with the results from morphology, physiological and biochemical characteristics,chemotaxonomic and phylegentic analysis based on 16S rRNA gene sequence studies, strain Xylan009 belonged to a yet unknown specie of the genus Amycolatopsis.
