摘要
水提醇沉玉米须得到的粗多糖类物质,经脱蛋白脱色及凝胶色谱纯化后分别经液相色谱和气相色谱测定,研究中分别考察了Sevag法和TCA法脱蛋白效果,交联葡聚糖凝胶色谱G-75和G-100对玉米须多糖的分离效果。结果表明,玉米须多糖经过反胶束溶液脱色,脱色率达到91·43%,多糖回收率达到77·59%。Sephadex G-100对玉米须多糖分离效果较好。玉米须多糖通过高效液相色谱鉴定玉米须多糖的纯度,无单一对称峰出现,表明多糖类物质不纯。通过气相色谱测定了玉米须多糖的单糖组成为甘露糖、半乳糖、木糖、阿拉伯糖和果糖五种单糖。
In the present work,the Stigma Maydis polysaccharides were obtained by water extraction and ethanol precipitation.Comparing the deproteinization effect of polysaccharides by Sevag with TCA.The polysaccharides were decolorized by reverse micelle solution, the polysaccharides decolorization ratio was 91.43% and the polysaccharides recovery was 77.59%.The purification of polysaccharides was obtained by SephadexG-100.By the HPLC identifying, there were some other objects except polysaccharides in the Stigma Maydis.The monosaccharide composition of the polysaccharides were mannose, galactose, xylose, fructose, arabinose.There were some different with other papers published.
