重组人类肝细胞生长因子对胶质瘤C6细胞垂体瘤转化基因的影响

摘要 目的探讨重组人类肝细胞生长因子(rhHGF)对胶质瘤细胞垂体瘤转化基因(PTTG)的影响。方法体外培养胶质瘤C6细胞,给予不同浓度的rhHGF(0、10、20、30 μg/L),半定量逆转录聚合酶链式反应(RT-PCR)检测PTTGmRNA表达,Western blot检测PTTG蛋白表达。结果不同浓度rhHGF作用后C6细胞PTTG mRNA表达与蛋白水平均增高,且随着浓度的增加,其升高越明显,各组间差异有统计学意义(P<0.01)。结论 rhHGF可上调PTTG的表达,且呈剂量依赖性。 Objective To investigate the effects of recombinant human hepatocyte growth factor (rhHGF) on pituitary tumor transforming gene (PTTG) in C6 glioma cells. Methods Glioma C6 cells were divided into control group and rhHGF treatment groups with rhHGF at 0,10,20,30 μg/L for 24 hours. After treatment, PTTGmRNA were detected by reverse transcription polymerase chain reaction (RT-PCR) and western blot was used to detected the expressions of PTTG protein. Results The expression levels of PTFG mRNA in all groups showed there were significant differences between each groups ( P 〈 0. 01 ). Significant differences were also found in PTTG protein expressions between each groups of C6 cells after treatment ( P 〈 0.01 ). Conclusion rbHGF can upregulate PTTG expression at both the mRNA and protein levels in glioma C6 cells with a dose-dependence manner.
