透明质酸酶在弓形虫重组融合抗原ROP2-P30基因免疫中的免疫增强作用 被引量:1

The role of immunoenhancement of hyaluronidase in DNA immunization with recombinant ROP2-P30 from Toxoplasma gondii

摘要 目的:研究透明质酸酶在弓形虫重组融合抗原ROP2-P30基因免疫中的免疫增强作用,初步探讨透明质酸酶在提高质粒DNA分子免疫效应中的应用潜能。方法:弓形虫重组质粒pcROP2-P30联合透明质酸酶以及单独的重组质粒pcROP2-P30肌肉注射免疫BALB/c雌性小鼠,生理盐水和空质粒载体pcDNA3.1作阴性对照免疫,免疫3次,间隔3周。免疫结束后,以纯化的弓形虫重组蛋白ROP2-P30(rROP2-P30)为检测抗原,检测各免疫组小鼠血清中特异性抗体的形成;cell counting kit-8(CCK-8)检测免疫鼠脾细胞的增殖活性;硝酸还原酶法检测免疫鼠脾细胞培养上清中NO的含量。结果:pcROP2-P30+透明质酸酶免疫组特异性抗体生成水平显著高于其它免疫组(均P<0.01);pcROP2-P30+透明质酸酶免疫组小鼠脾细胞受特异性抗原rROP2-P30刺激后,其淋巴细胞的增殖明显高于单独的质粒pcROP2-P30免疫组(P<0.01);pcROP2-P30+透明质酸酶免疫组脾细胞培养上清中免疫活性分子NO的含量也显著升高(均P<0.01)。结论:透明质酸酶在提高弓形虫DNA免疫效应方面显示了免疫增强作用,具有发展成为基因免疫的免疫佐剂的潜能。
