摘要
目的:观察2型单纯疱疹病毒对人永生化角质形成细胞株分泌IL-10、IFN-γ的影响,探讨角质形成细胞对HSV-2病毒感染初期的免疫防御机制。方法:用2型单纯疱疹病毒感染人永生化角质形成细胞,采用荧光实时定量PCR检测感染后24,48,72h的IL-10、IFN-γmRNA的表达量。结果:与对照组比较,经2型单纯疱疹病毒感染后人永生化角质形成细胞分泌IL-10的表达量在24h内下降,24~48h上升,48~72h下降,而IFN-γ则相反,24h内上升,24~48h下降,48~72h上升,与对照组比较差异均有统计学意义(P<0.05)。结论:在2型单纯疱疹病毒感染角质形成细胞初期,角质形成细胞具有类淋巴细胞的功能,能分泌IL-10、IFN-γ,在不同的时段产生不同的免疫反应,并且IL-10、IFN-γ的表达互为抑制。
Objective To investigate the role of HSV-2 on IL-10 and IFN-γ expression in HaCaT,and to explore the mechanism how human keratinocyte exerts immune defensive function upon HSV-2 infection.Methods HSV-2 was amplified in VERO cells.HaCaT cells were infected with HSV-2,and were harvested in 24,48 and 72 hours after infection.The expression levels of IL-10 and IFN-γ were detected with Real-time PCR.Results The expressions of IL-10 and IFN-γ were detected in normal HaCaT cells.After HSV-2 infection,expression of IL-10 decreased in 24 hours,increased in 48 hours and decreased again in 72 hours.In contrast,expression of IFN-γ was constantly higher in the HSV-2 infected groups than that in control group.Conclusion Upon HSV-2 infection,keratinocyte plays a lymphocyte-like role by secreting IL-10 and IFN-γ.And the expressions of IL-10 and IFN-γ change at different time points after HSV-2 infection.
