摘要
目的:观察热淋清颗粒对自身免疫性前列腺炎大鼠脾细胞增殖活性及前列腺组织炎性因子分泌的影响,进一步探讨其治疗慢性前列腺炎的可能作用机制。方法:采用大鼠前列腺蛋白提纯液辅以免疫佐剂造成实验性自身免疫性前列腺炎(EAP)大鼠模型,随机分为两组:治疗组给予热淋清颗粒原料药头花蓼浸膏2 g/kg;模型组给予相同体积的蒸馏水,另选一组正常组做对照。以上各组均灌胃给药,每天1次,连续给药30天。观察热淋清颗粒对各组大鼠脾细胞在刀豆蛋白A(ConA)、脂多糖(LPS)诱导下T、B淋巴细胞增殖活性的影响;采用放射免疫法检测分析热淋清颗粒对各组大鼠前列腺组织白细胞介素-1(IL-1)、IL-8、TNF-α及白介素-10(IL-10)等细胞因子分泌的影响。结果:热淋清颗粒能抑制脾淋巴细胞的增殖活性,抑制促炎细胞因子IL-1、IL-8及TNF-α的分泌,调节抗炎细胞因子IL-10的表达。结论:热淋清颗粒通过调节自身免疫性前列腺炎大鼠的免疫功能和促炎及抗炎因子的分泌而发挥治疗作用。
Objective:To study the effect of Relinqing Granules on splenocyte proliferative activity and inflammatory factor secretion of prostate tissues in rats with experimental autoimmune prostatitis in order to further approach the possible therapeutic mechanisms of Relinqing Granules on chronic prostatitis. Methods:The experimen tal autoimmune prostatitis (EAP) rat model was established by rat prostatein purification of FCA emulsion multiple intra-dermal injections. The model rats were randomly divided into model control group and treatment group, which were given Relinqing Granules(Touhualiao 2 g/kg) or distilled water respectively, and normal rats were taken as control group. The above groups were successively administrated orally for 30days, qd. The proliferation activity of T,B Lymphocytes induced by canavaline A and lipopolysaccharide in splenocytes was observed. Radioimmunoassy(RIA) was applied to detect the expression of IL-1, IL-8, TNF-α and IL-10 in the prostate tisuses. Results: Relinqing Granules could restrain proliferative activity of spleen lymphocyte, decrease the secretion of proinflammatory cytokines IL-1, IL-8 and TNF-a, and regulate the expression of anti-inflammatory cytokine IL-10. Conclusions:Relinqing Granules could regulate the immunological function, and the secretion of pro-inflammatory and anti-inflammatory cytokines to exert theraputic action on autoimmunue prostatitis.
