摘要
通过对5株杂交瘤细胞株分泌稳定性、抗体亚型、亲和力、特异性等方面的测定,表明其染色体众数为87~90条,具有众数的细胞所占比例为81%~84%,单抗亚型均为IgG1型,腹水单抗的亲和常数为108~109M-1,氟罗沙星单抗与诺氟沙星、环丙沙星、恩诺沙星、链霉素、庆大霉素、卡那霉素的交叉反应试验表明,所制备的抗氟罗沙星单抗对氟喹诺酮类药物有较高的交叉反应率,与链霉素、庆大霉素、卡那霉素无交叉反应。
The quality of the prepared anti-Fleroxacin monoclonal antibodies were evaluated by the stability of secretion, antibody-subtype, affinity and specificity experiments' results. It showed that the five strains of hybridoma cells contained modal number of chromatin for 87-90 which occupied for 81%-84%, subtypes of monoclonal antibodies were all IgG1 types, affinity constant of aseites monoclonal antibodys was 10^8-10^9M^-1. The cross-reaction experiments of Fleroxacin monoclonal antibody were carried out with Norfloxacin, Ciprofloxacin, Enrofloxacin, Streptomycin, Gentamicin, Kanamycin. From the experiments we know that the prepared anti-fleroxacin monoclonal antibodies had a very high reaction rate with fluoroquinolones, but had no reaction with streptomycin, gentamiein, kanamyein.
