摘要
探讨细胞因子和地塞米松对肾小球系膜细胞表面表达细胞间粘附分子1(ICAM1)的影响。通过体外系膜细胞培养的方法,分别加入不同的细胞因子,观察其浓度及作用时间产生的影响;并用ELISA法检测不同浓度的地塞米松对系膜细胞表达ICAM1的影响。结果显示系膜细胞表达ICAM1的强弱与细胞因子的作用呈剂量、时间依赖关系;随着地塞米松浓度的增加,ICAM1表达逐渐减弱,与未加地塞米松组相比相差均非常显著(P<001),表明细胞因子促使系膜细胞表达ICAM1增强,而地塞米松抑制系膜细胞表达ICAM1,这对进一步阐明肾小球炎性疾病的发病机理及选择、指导临床用药具有参考意义。
To study the effect of expressing intercellular adhesion molecule1 on glomerular mesangial cell by cytokines and dexamethasone.Mesangial cells were cultured in vitro and different cytokines were added.The effects of its dose and time were observed.Changes of ICAM1 expressing on mesangial cell were evaluated by ELISA with adding different dosages of dexamethasone.It was found that the intensity of expressing ICAM1 on mesangial cell was dose and timedependent with cytokines.ICAM1 expressing was attenuating with increase of dosages of dexamethesone.The difference was significant compared with control group(P<001).The results indicated that intensity of expressing ICAM1 on mesangial cell was enhanced by cytokines.ICAM1 expressing was inhibited by dexamethasone.It will elaborate the pathogenesis of glomerular inflammation and guide chosing clinic drugs.
