摘要
目的:建立测定青叶胆中獐牙菜苦苷和异牡荆素含量的高效毛细管电泳分析方法。方法:青叶胆药材粉末经50%甲醇溶液超声提取后,采用未涂层弹性石英毛细管柱(60.2cm×75μm,有效长度50cm)作为分离通道;以含5%甲醇的50mmol·L-l硼砂(pH=9.37)为运行缓冲溶液;分离电压15kV;检测波长236nm;柱温25℃。结果:獐牙菜苦苷和异牡荆素浓度分别在0.1304~1.304mg·mL-l(r=0.9998)和0.0298~0.298mg·mL-l(r=0.9994)范围内呈良好的线性关系;平均回收率(n=6)分别为98.5%和99.3%,RSD均小于2%。结论:本法简便、快速,可用于青叶胆中獐牙菜苦苷和异牡荆素的含量测定。
Objective:To develop an HPCE method for determination of swertiamarin and isovitexin in Swertia mileensis T.N.Ho et W.L.Shih.Methods:Swertiamarin and isovitexin in Swertia mileensis were extracted with 50% methanol solution by supersonic wave technique,and then determined by capillary electrophoresis.A uncoated fused-silica capillary column (60.2 cm×75 μm,an effective length of 50 cm) was adopted.The running buffer was composed of 50 mmol·L-l borax solution containing 5% methanol (pH=9.37).The applied voltage was 15 kV and the capillary temperature was 25 ℃.The detection wave length was 236 nm.Results:A good linearity between peak area and concentration was found in the range of 0.1304-1.304 mg·mL-l(r=0.9998) for swertiamarin and 0.0298-0.298 mg·mL-l(r=0.9994) for isovitexin;Their average recoveries(n=6) were 98.5% and 99.3% respectively,and the relative standard deviation was less than 2.0%.Conclusion:The method is simple and rapid.It can be used to determine swertiamarin and isovitexin in Swertia mileensis.
