摘要
目的:采用高效液相色谱-电喷雾离子阱质谱(HPLC-MSn),分离和鉴定磷霉素氨丁三醇及其有关物质。方法:色谱柱为Synergi Hydro-RP C18柱(150mm×2.0mm,4μm);流动相为10mmol·L-1正辛胺溶液(用冰醋酸调节pH5.2)-乙腈,梯度洗脱;流速0.3mL·min-1;柱温35℃;在负离子模式下,用电喷雾离子阱质谱法对磷霉素及其有关物质进行多级质谱分析。结果:分离了磷霉素及其5种有关物质,并对分离的杂质进行结构确证。
Objective:To develop an HPLC with ESI-ion trap mass spectrometry detection method for separation and identification of fosfomycin trometamol and its related substances.Method:The separation was performed on a Synergi Hydro-RP C18 column(150 mm×2.0 mm,4 μm) with optimized mobile phase of 10 mmol·L-1 octylamine solution (adjusted to pH 5.2 with glacial acetic acid)-acetonitrile gradient elution at a flow rate of 0.3 mL·min-1.The column temperature was 35 ℃.Under the negative ion mode,utilizing ESI-ion trap mass spectrometry analysis fosfomycin trometamol and its related substances.Result:Succesfully separated and identified fosfomycin trometamol and its related substances.
