摘要
目的:通过实验,建立一种测定穿琥宁的新方法。方法:在pH4.5的酸性溶液中,取适量穿琥宁溶液和伊文思蓝溶液混合,以二次蒸馏水稀释,放置一段时间后在荧光光度计上以λem=λex方式进行同步扫描。结果:伊文思蓝和穿琥宁能够迅速反应生成离子缔合物,导致共振瑞利散射(RRS)光谱的改变,产生比较特征的新的RRS光谱,最大峰值在717nm,并且在25~900mg·L-1范围内,RRS强度和穿琥宁的浓度成正比。结论:我们建立了测定穿琥宁的共振瑞利散射法。该法灵敏度较高,检测限可达到45ng·mL-1,用于检测某些穿琥宁样品中穿琥宁的含量,结果较满意。
Objiective:We wish to found a method to determine PDS in medicine.Method:In pH 4.5 HAc-NaAc butter solution,potassium dehydroandrographolide succinate (PDS),water and evans blue (EB) with proper properation were mixed.After a while,it was put in Hitachi F-2500 and determined the resonance rayleigh scattering(RRS) spectra.Result:EB reacts rapidly with PDS to form complexes,which results in great enhancement of resonance rayleigh scattering(RRS)and a new RRS spectrum.The maximum RRS peak was near 717 nm.RRS intensity was proportional to the concentration of PDS in the range of 25-900 mg·L-1.Conclusion:A method can be used to determine PDS based on our experiments.The method has high sensitivity and the detect limit (3σ) for PDS is 45 ng·mL-1.The method has been used for the determination of total amounts of PDS in samples with satisfactory results.
