摘要
对酶法提取侧柏叶总黄酮类的工艺条件进行研究。以总黄酮得率为考察指标,探讨了影响提取的主要因素,并用正交实验确定了最佳提取工艺。结果表明,各因素对提取效果影响的顺序依次为酶解时间>酶用量>酶解温度>酶解pH值,最佳工艺条件为:料液比1∶14,酶解温度45℃,介质pH3.5,酶用量0.3%,酶解时间2.0h,总黄酮得率可达1.026%。该提取工艺简便易行,重复性好。
To optimize the extraction technique of total flavonoids from Platycladus orientalis leaves by enzymatic hydrolysis. The main influential factors were studied and the optimum extraction technology was obtained by orthogonal test guided by total flavonoids yield. Results: The results revealed that the factors influencing the extracting process was in the order of hydrolysis time〉 enzyme dosage〉 hydrolysis temperature〉pH value, and the optimum extracting technology was as follows: solid-liquid ratio was 1 : 14, enzymatic hydrolysis temperature was 45 ℃, enzyme dosage was 0. 3%, pH value was 3.5, treatment time was 2. Oh. The total flavonoids yield was as high as 1. 026%. The optimum extraction technique is simple to conduct and has good repeatability.
