摘要
目的探讨促红细胞生成素(EPO)对脑缺血再灌注(IR)大鼠海马CA1区Bcl-xl表达和认知功能的影响。方法将雄性SD大鼠随机分为假手术组、IR组、EPO组,并制作IR模型;制模前3h,EPO组及IR组大鼠脑室立体定向分别注射重组人EPO(rHu-EPO)及生理盐水。制模24h后,应用免疫组化法检测各组大鼠海马CA1区Bcl-xl蛋白表达;4周时电迷宫测验各组大鼠学会逃避及记忆再现所需的电击次数。结果与假手术组比较,制模24h时,EPO组和IR组海马CA1区Bcl-xl蛋白表达明显减少,但EPO组明显多于IR组(均P<0.01);制模4周后,EPO组及IR组大鼠学会逃避所需的电击次数明显增多,但EPO组明显少于IR组(均P<0.01);EPO组及IR组大鼠记忆再现次数明显减少,但EPO组多于IR组(均P<0.01)。结论EPO预处理可以促进IR大鼠海马CA1区Bcl-xl蛋白表达,改善IR大鼠的认知功能损害。
Objective To explore the effect of erythropoietin(EPO) on Bcl-xl expression in hippocampal CAI subregion and cognizing function in rats with cerebral ischemia-reperfusion ( IR ). Methods Male SD rats were randomly separated into sham-operated group, IR group and EPO group, and the IR models were made. At 3 h before IR models made, rHu:EPO and saline were injected into rat lateral ventricles in EPO group and IR group, respectively., with help of stereotaxic coordinates. Bcl-xl protein expression of Hippocampal CA1 subregion was detected by immunohistochemistry at 24 h after operation. The learning and memory abilities were examined by electricity maze at 4 weeks after operation. Results Compared to sham-operated group, Bcl-xl protein expression in EPO group and IR group were obviously decreased, but EPO group was significantly increased than IR group( all P 〈 0. 01 ). After 4 weeks, the stroke times of learn escaping in EPO group and IR group were more than sham-operated group,but EPO group was less than IR group ( all P 〈 0. 01 ) ; EPO group and IR group had less memory emersion times than sham-operated group,but EPO group had more memory emersion times than IR group (all P 〈 0. 01 ). Conclusion EPO pretreatment can enhance the expression of Bcl-xl protein in hippocampal CAI subregion and improve cognizing disfunction of IR rat.
