摘要
目的分析小眼虫细胞Z株和SMZ株对二氯化镉的毒性反应特点。方法采用Koren-Hutner细胞培养液培养小眼虫细胞,制成细胞悬液接种于96孔培养板,二氯化镉染毒,浓度为6.25-800.0μM。于染毒培养后的24h和48h,用FLUOstarOPTIMA多功能微板读取仪分别于波长610nm和490nm处读取小眼虫细胞吸光度(ABS)和细胞增殖活性(MTS),光镜下观察形态学变化。结果二氯化镉浓度为6.25μM和12.5μM时即可对小眼虫细胞的SMZ和Z株细胞的增殖有明显的毒性作用,与对照组比较差异有统计学意义(P<0.05),对Z株细胞24、48h后的ABS及24h低于200μM组的MTS检测未见明显改变,与对照组比较差异无统计学意义;染毒后48h光镜下观察小眼虫细胞Z株和SMZ株均可见V形、星形及多形细胞。结论小眼虫细胞对受试物浓度变化的反应是SMZ株较Z株敏感,Z株细胞对二氯化镉显示一定程度的耐受。
Objective To analyze the poisonous effects of CdCl2 against Euglena gracilis,both strains Z and SMZ under continual exposure.Methods All experiments were performed with Euglena gracilis Z and SMZ,preliminary grown in Koren-Hutner medium until reaching the early stationary.Cells were inoculated in 96-well culture plates with a CdCl2 concentration of 6.25μM-800.0μM.To check the absorbance (ABS) and viability (MTS) at the wavelength of 610nm and 490 nm with FLUOstar OPTIMA spectrometer,to observe the abnormal cells with microscope with CCD camera after 24h and 48hs incubation treated with CdCl2Results The poisonous effects of CdCl2 to two strains SMZ and Z of E.gracilis at a concentration above 6.25 μM and 12.5 μM were obvious compared with controls (P0.05),the viability (MTS) and absorbance (ABS) were detected and no difference compared with control after 24h and 48hs to Z lines,24h to SMZ line less than 200 μM group.More abnormal cells (V-shape,Starfish-shape hypertrophy) were observed under the microscope with CCD camera in both stains at different levels of CdCl2.Conclusion The response of Strain SMZ to CdCl2 is more sensitive than that of Z,and Z strain shows more resistance.
