细粒棘球绦虫重组Bb-Eg95-EgA31疫苗免疫小鼠后脾细胞增殖的动态观察 被引量:1

Dynamic observation of splenocyte proliferation in mice immunized with a recombinant Bb-Eg95-EgA31 vaccine of Echinococcus granulosus

摘要 目的动态观察细粒棘球绦虫(Eg)重组双歧杆菌(Bb)-Eg95-EgA31疫苗免疫小鼠后脾淋巴细胞增殖的变化。方法将5×108克隆形成单位(CFU)和5×105CFU疫苗分别采用口服灌胃和鼻腔内接种方法免疫BALB/c鼠,免疫后0、2、4、6、8、10、12、14、16、18和20周各剖杀4只小鼠,取脾,分离脾细胞,用Eg粗抗原(EgAg)或刀豆素A(ConA)刺激培养,四甲基偶氮唑盐比色法(MTT法)检测免疫小鼠脾淋巴细胞增殖反应,同时设有MRS对照。结果口服灌胃组脾淋巴细胞增殖水平在免疫后410周升高,在免疫后6周达最高水平;鼻腔内接种组脾淋巴细胞增殖水平在免疫后48周升高,在免疫后6周达最高水平。结论细粒棘球绦虫重组Bb-Eg95-EgA31疫苗可诱导小鼠脾淋巴细胞增殖。 Objective To dynamically observe changes in splenocyte proliferation in mice immunized with a recombinant bifidobacteria bifidum(Bb)-Eg95-EgA31 vaccine of Echinococcus granulosus(Eg).Methods BALB/c mice were vaccinated with 5×108 colony forming units(CFU) orally and 5×105 CFU intranasally.Mice were sacrificed in weeks 0,2,4,6,8,10,12,14,16,18,and 20 after immunization and their spleens were isolated for cell culture and cell proliferation assay following stimulation with EgAg or concanavalin A(ConA).The proliferation and viability of splenocytes was measured using the methyltetrazolium(MTT) assay,with MRS as a control.Results In the orally immunized group,the level of splenocyte proliferation significantly increased during the 4-10th week after vaccination and peaked in the 6th week after vaccination;in the intranasally immunized group,the level of splenocyte proliferation clearly increased in the 4-8th week after vaccination and peaked in the 6th week after vaccination.Conclusion Prol...更多iferative activity of splenocytes can be induced in mice immunized with a recombinant Bb-Eg95-EgA31 vaccine of E.granulosus.
