Genetype Characteristics of Enterovirus Type 71 in Shenzhen from 2005 to 2008

摘要 对2005-2008年深圳地区肠道病毒71型进行基因特征研究。用肠道病毒71型特异性引物进行RT-PCR,并扩增EV71的VP1基因,所得的核苷酸序列与肠道病毒71型A、B、C基因型代表株的核苷酸序列比较,并且用DNASTAR、BioEdit和Mega 3.1软件进行系统进化分析。结果表明17株病毒与C4a亚群代表株接近,VP1区核苷酸同源性在95.3%-99.4%之间,1株病毒与C4b亚群代表株较接近,核苷酸同源性在93.0%-95.6%之间;18株病毒与A、B基因型代表株比较差异较大,VP1区核苷酸同源性为81.8%-86.0%,18株病毒之间的VP1核苷酸同源性为92.5%-100%。深圳地区EV71的流行株均属于C4基因型,但以C4a亚群为主,还存在C4b亚群。 Genetic characteristics of enterovirus type 71 in Shenzhen from 2005 to 2008 were analyzed. All samples were detected by RT-PCR using EV71-specific primers. The VPls of EV71 strains were amplified and sequenced. A phylogenetic tree was constructed by comparison of the sequences with those of subgenotype A, B and C using DNASTAR, BioEdit and Mega 3.1 softwares. The VP1 nucleotide sequences of 17 strains of Shenzhen shared 95.3%-99.4% identities with cluster C4a, and one strain shared 93.0%- 95.6% identities with cluster C4b. The homogeneity of the VP1 nucleotide sequence with subgenotype A and B was 81.8%-86.0%. Among 18 strains, the homogeneity of the VP1 nucleotide sequence was between 92.5%-100%. All EV71 strains circulating in Shenzhen were of subgenotype C4. The predominant strain of EVT1 belonged to cluster C4a, also there was cluster C4b.
