胃癌组织中标记滞留细胞的初步鉴定

摘要 目的研究裸鼠胃癌组织中标记滞留细胞的存在及其分布特点。方法培养人胃癌细胞株BGC-823,5-BrdU标注后接种于裸鼠右腋窝皮下。于接种后10、20、28、35、42、49、56d处死裸鼠,取其右腋下肿瘤结节进行免疫组化检测。结果实验组接种后第10天,裸鼠肿瘤组织中标记滞留细胞(LRCs)表达最高,达到96.14%,随着时间的增加,LRCs逐渐降低,到第80天在肿瘤中仅有极少量细胞表达BrdU,约占0.51%,主要位于癌巢的边缘部位,细胞形态与BrdU阴性细胞无明显的差异。结论胃癌BGC-823细胞株在裸鼠体内形成的肿瘤内存在标记滞留细胞,这部分细胞与肿瘤干细胞密切相关,可能在维持肿瘤生长、转移、复发方面发挥重要作用。 Objective To study the existence of label-retaining cell and its distribution in gastric cancer of nude mouse.Methods The gastric carcimoma cell line BGC-823 was labeled with BrdU in vitro and then engrafted into the right oxter of nude mice,which developed tumors.Label-retaining cells(LRCs) were quantified by immunohistochemical methods.Results BrdU positive cells constituted about 96% of the cells in xenograft tumors after 10 days.Subsequently,BrdU positive cells graduatedly decreased,at the 56 days LRCs steadily occupied about 0.5%,this set of population cell localized in the margin of cancer nests,which had no difference in cellular morpha.Conclusion This study has demonstrated for the first time the presence of LRCs in human gastric cancer xenografts in nude mice and the LRCs may be related with cancer stem cells,which are most likely the cause for spread,metastasis and recurrence.
