摘要
目的研究大鼠隐睾睾丸组织生精功能,了解睾丸生殖细胞凋亡。方法建立隐睾大鼠动物模型,获得单侧隐睾大鼠(A组)12只,双侧隐睾大鼠(B组)10只,空白对照正常大鼠(C组)10只。当日龄为60d时,切取大鼠双侧睾丸组织,行HE染色、Johnsen评分;采用原位缺口末端标记法检测生殖细胞凋亡。结果A、B组隐睾睾丸组织Johnsen评分较对照组显著降低(P〈0.05),生精功能明显破坏,生殖细胞凋亡指数显著增加(P〈0.05)。结论大鼠隐睾睾丸组织生精功能受到损害,生殖细胞凋亡增加。
Objective To study the impairment of spermatogenesis function and the apoptosis of reproductive cell in rat crytorchidism testicle. Methods Thirty two male Sprague Dawley rats were divided into 3 groups:Group A (n = 12),unilateral cryptorchidism; Group B (n = 10),bilateral cryptorchidism, and Group C (n = 10) ,control group. Group A and B obtained from the cryptorchidism model was induced by 17-β estradiol. Estimation of spermatogenesis by application of the Johnsen score revealed impairment of germinal cell maturation at day 60 postpartum. Germ cell apoptosis was detected by TdT-mediated dUTP-X nick end labeling in three groups. Result Compared with group C, the impairment of spermatoge-nesis function of testicular tissue of each cryptorchidism in group A and B were obviously ( P 〈 0. 05 ) , Germ cell apoptosis index increased significantly (P 〈 0. 05 ). Conclusion The spermatogen- esis function of cryptorchidism testicular tissue in rats was damage, the increase of reproductive cell apoptosis associated with the damage.
