地衣芽孢杆菌1411-1羽毛角蛋白固体发酵工艺的优化及角蛋白酶的初步研究 被引量:4

摘要 对角蛋白高效降解菌地衣芽孢杆菌菌株1411-1固体发酵条件进行了优化,着重考察了培养基含水量、起始pH值、温度、接种量及外源添加物的影响,并确定了优化的发酵周期,同时对角蛋白酶的提取进行了初步研究。研究结果表明,1411-1的最适固体发酵条件为:培养基含水量75%,pH值10.0,温度40℃,菌液接种量0.15ml/g,发酵周期88h,氮源对固体发酵有显著的抑制作用,碳源的影响不显著;角蛋白酶的最适提取条件为:以碳酸钠-碳酸氢钠缓冲液(pH值10.0)作为提取剂,4℃下提取60min。
