摘要
本文首次利用FT—IR光谱技术和计算机辅助解析法(二阶导数、去卷积和曲线拟合)研究了天花粉蛋白重水溶液(pD6.5)中的二级结构。研究结果表明:室温下,天花粉蛋白溶液中含43%的α—螺旋、24%的β—折叠和33%的无规结构。这与晶体衍射方法得到的结果完全吻合。
Fourier-Transform infrared(FT-IR) spectroscopy is a valuable method for the study of protein conformation in solution. Combined with resolution-enhancement techniques such as second-derivative spectroscopy and Fourier self-deconvolution, plus the application of the curve fitting technique , FT-IR was used to test for the presence of secondary structures of TCS in D2O. The result shows that the contents of secondary structures of TCS at room temperature are as followings : α-helix 43%, β-sheet 24%, random structure 33%. This result is consistent with that of x-ray experiment.
