摘要 通过观察医用硅橡胶对成骨细胞的骨桥蛋白mRNA基因表达的影响,进行生物材料的分子生物相容性研究方法的初探.将医用硅橡胶的浸提液与原代成骨细胞共同培养,用骨桥蛋白探针与细胞进行原位杂交.镜下观察细胞内有无特殊的染色颗粒.结果表明:无细胞毒性的整形外科用植入性硅橡胶材料组显示出该基因表达的蓝黑色颗粒;有细胞毒性的口腔印模硅橡胶则无明显表达现象.原位杂交法观察医用硅橡胶对成骨细胞骨桥蛋白基因表达的影响,可作为评价生物材料的分子生物相容性的参考,将可用于对骨植入材料和组织工程材料的分子生物相容性研究. To observe medical silicone rubber on osteopontin mRNA gene expression in osteoblast and evaluate the molecular biocompatibility. Extracted soluition of medical silicone rubber were diluted 10% solution and were co-cultured with the primary SD rat cranium cells for 5days in vitro. Cells in situ hybridization was done using the digoxin labeling osteopontin probe. The negative control was the normal saline and positive control was the phenol. Silicone rubber for the plastic surgery that no cytotoxicity , was showed the blue - black grain of that gene expression. While, silicone rubber for oral impression of dentistry that had the serious cytotoxicity, was showed no the grain of osteopontin gene expression. Effect of medical silicone rubber on osteopontin gene expression in primary cultured osteoblast was observed by cell in situ hybridization. It can be used for investigating the molecular biocompatibility especially on bone implanted materials and materials for tissue engineering.
