The effects of raloxifene and estradiol benzoate on expression of estrogen sulfotransferase in human breast tissues implanted into nude mice

摘要 目的观察雷洛昔芬和苯甲酸雌二醇对植入裸鼠的人乳腺组织雌激素硫酸转移酶(EST)表达的影响。方法15例正常乳腺组织植入75只裸鼠。每例标本分5份,分别植入5只裸鼠后分为5组:对照(Ⅰ组)、苯甲酸雌二醇1 mg/kg(Ⅱ组)及雷洛昔芬0.3 mg/kg(Ⅲ组)、3 mg/kg(Ⅳ组)、30 mg/kg(Ⅴ组)。给药30 d后取出移植的乳腺组织,采用逆转录聚合酶链反应(RT-PCR)方法检测EST mRNA的表达。结果与Ⅰ组相比,Ⅱ组EST mRNA表达升高;半定量EST mRNA约为对照组的2倍(P<0.05)。Ⅲ、Ⅳ、Ⅴ三组EST mRNA表达与Ⅰ相比无显著性差异(P>0.05)。结论剂量1mg/kg的雌激素上调EST表达,雷洛昔芬对正常乳腺的EST表达无影响。 Objective: To investigate the effect of raloxifene and estradiol benzoate on expression of estrogen sulfotransferase (EST) in normal human breast tissue implanted into nude mice. Methods: Fifteen specimens of normal human breast tissues from patients with benign breast disease were implanted into 9-10-week-old athymic nude mice. Every specimen was divided into five parts and individually implanted into five mice. The mice were divided into 5 groups subjected to the following treatments: distilled water (Controls, Group Ⅰ ); estradiol benzoate (B-E2) at 1 mg/kg (Group Ⅱ ); raloxirene at 0.3 mg/kg (GroupⅢ), 3 mg/kg (Group Ⅳ) and 30 mg/kg (GroupⅤ). Treatment was given every day, and human breast tissues were removed for animals after treatment for 30 days. The expression of EST mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR). Regults: The semi-quantitative EST mRNA increased by about twofold in breast tissue in estradiol benzoate group compared to control (PM0.05), but there was no significance in every group of raloxifene compared with control (P〉0.05). Conclusions: Dose of 1 mg/kg estradiol benzoate can regulate up the EST expression, while raloxifene has no effect on EST of normal breast tissues.
