摘要
目的:探讨高迁移率族蛋白(high mobilitygroup protein box1,HMGB1)对结肠癌细胞血管内皮生长因子C(vascular endothelia growth factor C,VEGF-C)的表达影响及核转录因子(nuclear factorκB,NF-κB)在其中的作用。方法:以HMGB1作为干预因素,通过RT-PCR方法检测在HMGB1作用不同时间后,结肠癌细胞HCT116中VEGF-C表达的变化。构建NF-κB干扰质粒,检测比较干扰前后HMGB1对结肠癌细胞VEGF-C表达影响的变化。结果:HMGB1可诱导HCT116细胞中VEGF-C的表达,随着对结肠癌细胞作用时间的延长,VEGF-C的表达逐渐增强,在2、4、8、12和24h几个时间点中,以12h时HMGB1对VEGF-C的表达影响最明显。抑制NF-κB可削弱HMGB1对VEGF-C的诱导作用。结论:结肠癌细胞中HMGB1可通过刺激NF-κB活化的途径,诱导VEGF-C。
OBJECTIVE:To explore the secretion pattern of VEGF-C in HCT116 cells induced by HMGB1 and the function of NF-κB in this progress.METHODS:Six groups of HCT116 were cultured with the same concentration of HMGB1 for different time.The changes of VEGF-C expression were detected by RT-PCR at every time point.The role of NF-κB in the progress was analyses by RNA interference.RESULTS:HMGB1 induced VEGF-C expression in HCT116 cells.VEGF-C expression increased after HMGB1 was added into the culture solution.Among the 6 time points(2,4,8,12 and 24 h),HMGB1's effect on the expression of VEGF-C reached the peak value at 12 h.The inhibition of NF-κB diminished the effect of HMGB1 on VEGF-C.CONCLUSION:HMGB1 can induce the expression of VEGF-C in colon cancer through NF-κB pathway.
