Research progress of Smad4 in common malignant tumor tissues of digestive system

摘要 目的:总结国内外关于Smad4在消化系统常见恶性肿瘤中研究进展。方法:应用PUBMED和CNKl期刊全文数据库检索系统,以"Smad4"和分别以"胰腺癌、食管癌、胃癌"等为关键词,检索1996-01-2009-01有关Smad4的文献205篇,其中英文文献160篇,中文文献45篇。纳入标准:1)Smad4的功能域、信号转导和调节;2)Smad4在消化系统常见恶性肿瘤中的表达情况;3)Smad4表达缺失与肿瘤进展相关性;4)以Smad4为靶点的肿瘤靶向研究。根据纳入标准,精选85篇文献,最后纳入分析25篇文献。结果:Smad4基因在胰腺癌,消化道腺癌中起着肿瘤抑制因子的作用,其表达缺失与这些肿瘤发生发展呈负相关,而在食管鳞癌中Smad4基因转录和其蛋白表达水平不一致且Smad4蛋白表达缺失与食管鳞癌的分级和分期的相关性研究文献较少。结论:深入研究Smad4在消化系统常见恶性肿瘤尤其是食管鳞癌中的表达及功能,有助于进一步理解肿瘤侵袭和转移的分子生物学机制,有望在肿瘤的靶向治疗方面取得新的突破。 OBJECTIVE:To summarize the research of Smad4 in common malignant tumors of the digestive system home and abroad.METHODS:By PUBMED and CNKI retrieval system,with"Smad4 and pancreatic cancer,esophageal squamous cell carcinoma,gastric cancer et al."as the key words,160 papers of literature in English and 45 in Chinese on Smad4 were retrieved from 01,1996 to 01,2009.The selection standards for the data related to SM-AD4:1)domain and function of signal transduction and regulation of Smad4;2)the expression of Smad4 in common malignant tumors of the digestive system;3)tumor progression associated with loss expression of Smad4;4)targeting Smad4 as an anticancer therapy.According to the above standards,85 documents were selected and 25 were finally analyzed.RESULTS:Smad4 gene as a tumor suppressor plays a crucial role in pancreatic cancer and gastrointestinal adenocarcinoma tumor,besides,the lack of the expression of Smad4 has a negative correlation with the development of these tumors.However,it is inconsistent that Smad4 gene transcription and its protein expression level,and relevant research literature few between loss expression of Smad4 protein and the grade and stage in esophageal squamous cell carcinoma.CONCLUSION:Further research on the expression and function of Smad4 in the common malignant tumors of the digestive system,especially in the esophageal squamous cell carcinoma will contribute to extensity understanding the molecularmechanism of tumor invasion and metastasis and acquire an important breakthrough in cancer targeted therapy.
