摘要
目的:检测Kangai-1(KAI-1)mRNA和基质金属蛋白酶-9(MMP-9)mRNA在胃癌组织中的表达,探讨其与胃癌临床病理特征之间的关系。方法:采用反转录-聚合酶链反应(RT-PCR)检测86例胃癌组织中KAI-1 mRNA、MMP-9 mRNA的表达。结果:KAI-1 mRNA在高分化腺癌组织中的表达率(58.82%)明显高于低分化者(15.56%)(P<0.05),MMP-9 mRNA表达率在高分化和低分化腺癌间差异无统计学意义(P>0.05);无淋巴结转移的胃癌组织中KAI-1 mRNA表达率(61.90%)明显高于有淋巴结转移者(23.08%)(P<0.05),MMP-9 mRNA则相反(P<0.05);KAI-1 mRNA表达率随浸润深度增加而逐渐降低,MMP-9则逐渐升高(P<0.05)。KAI-1 mRNA在TNMI期胃癌的表达率明显高于TNMⅢ期和Ⅳ期者(P<0.05),MMP-9差异则无统计学意义(P>0.05)。结论:KAI-1 mRNA在胃癌组织中的表达与肿瘤浸润深度、分化程度、TNM分期及淋巴结转移有关;MMP-9 mRNA在胃癌组织中的表达与肿瘤浸润深度、淋巴结转移有关。
Objective To determine the expressions of KAI-lmRNA and MMP-9 mRNA in gastric cancer and to study their relationship with clinical pathologic characteristics. Methods Expressions of KAI-I mRNA and MMP-9 mRNA in 86 gastric cancer specimens were assessed by reverse transcription polymerase chain reaction (RT-PCR). Results Expression rate of KAI-1 mRNA in well-differentiated gastric cancer (58.82%) was significantly higher than that of poorly differentiated gastric cancer (15.56%) (P〈0.05), while the expression rate of MMP-9 mRNA was not statistically different (P〉0.05).KAI-1 mRNA expression rate in gastric cancer without lymph node metastasis (61.90%) was significantly higher than that with lymph node metastasis (23.08%) (P〈0.05); MMP-9 mRNA expression rate was the reverse (P〈0.0g). The expression rate of KAI-1 mRNA decreased with the increase in depth of infiltration, while MMP-9 mRNA expression rate increased gradually (P〈0.05). The expression rate of KAI-1 mRNA in TNM Ⅰ stage was significantly higher than that in TNM Ⅲ/Ⅳ stage (P〈0.05); no statistically significant difference was seen in MMP-9 mRNA expression rate (P〉0.05). Conclusions The expression rate of KAI-1 mRNA in gastric cancer is correlated with depth of infiltration,degree of differentiation ,TNM staging and lymph node metastasis, while the MMP-9 mRNA expression rate was correlated only with depth of infiltration and lymph node metastasis.
