摘要
目的探讨氯化甲基汞(MMC)诱导NCI-H446细胞凋亡发生的可能机制。方法通过分子生物学方法,检测调控细胞凋亡的关键基因Bcl-2、Bax和caspase-3在mRNA水平的表达情况。结果MMC组细胞Bcl-2mRNA水平低于对照组,并存在着时间依赖性降低趋势,至24h达到最低;Bax及caspase-3mRNA水平高于对照组,并存在着时间依赖性增高趋势,至24h达到最高。结论MMC能够在体外诱导NCI-H446细胞发生凋亡。
Objective To investigate the possible mechanism of methylmercury (MMC) inducing apoptosis in human small cell lung cancer NCI-H446. Methods The expressions of Bcl-2, Bax and caspase-3 in NCI-Hd46 cells were detected by RT-PCR. Results The mRNA level of Bcl-2 in MMC group was lower than that of control group, and had time dependence, was the minimum at 24 h. The mRNA levels of Bax and caspase-3 in MMC group were higher than those of control group, and had time dependence, were the maximum at 24 h. Conclusions MMC could induce NCI-Hdd6 cells apoptosis in vitro.
