Determination of Allantoin in Xiao'er Jianpi Granules by HPLC

目的:采用HPLC法测定小儿健脾颗粒中尿囊素的含量。方法:采用ZORBAX SB-C_(18)(250 mm×4.6mm,5μm)色谱柱,以乙腈-0.005 mol·L^(-1)磷酸二氢钾(0.8:100)作为流动相,流速为0.8 ml·min^(-1),柱温:30℃,检测波长:224 nm。结果:尿囊素在21.5～430.0μg·ml^(-1)范围内有良好的线性关系,平均加样回收率为97.92%,RSD=1.12%。结论:本方法快速简便,结果准确,可用于该制剂中尿囊素的定量分析。

Objective： To estabish an HPLC method for the determination of allantoin in xiao＇ er jianpi granules. Method： The ZORBAX SB-Cls （250 mm×4.6 mm ,5 μm） column was used,the mobile phase consisted of acetonitrile ：0. 005 ml. Potassium dihydrogen phosphate （0.8：100 ,v/v） ,the flow rate was 0.8ml·min^-1 ,the column temperature was 30 ℃the detecting wavelength was at 224 nm. Result： The cablibration curve was linear within a range of 21.5-430.0 μg·ml^-1 , the average recovery of this method was 97.92% and the RSD was 1.12%. Conclusion： The method is simple, repeatable and accurate,it can be applied in quantitative determination of allantoin in xiao＇ er jianpi granules.
