摘要
目的:探讨茶多酚和银杏叶提取物对模拟失重所致的高血管紧张素Ⅱ(AngⅡ)状态刺激血管内皮细胞分泌内皮素的影响的保护作用。方法:将培养的血管内皮细胞分为6组:空白对照组,单纯AngⅡ组,AngⅡ+高、低浓度茶多酚组(研究Ⅰ、Ⅱ组),AngⅡ+高、低浓度银杏叶提取物组(研究Ⅲ、Ⅳ组),用放免法测定在不同时间作用下内皮细胞分泌内皮素含量的变化。结果:①AngⅡ有明显促进血管内皮细胞分泌内皮素的作用。②茶多酚和银杏叶提取物能明显抑制高AngⅡ致血管内皮细胞分泌内皮素的作用(P<0.01)。③低浓度茶多酚和低浓度银杏叶提取物较高浓度茶多酚和高浓度银杏叶提取物的作用更强(P<0.01)。结论:①高AngⅡ状态可明显刺激血管内皮细胞分泌内皮素。②低浓度茶多酚和银杏叶提取物可显著降低高AngⅡ状态所致的内皮素分泌增加,从而起到保护血管内皮细胞的作用,两者的生理作用无明显差异。
Objective:To observe the effect of Tea polyphenols and Ginaton on vascular endothelial cells(VEC) secreting endothelin (ET) stimulated by high level of Ang Ⅱ caused by simulated microgravity. Methods: Cultured VEC were divided into 6 groups:control group,Ang Ⅱ group,Ang Ⅱ + high concentration Tea polyphenols group,Ang Ⅱ + low concentration Tea polyphenols group,AngⅡ + high concentration Ginaton group and Ang Ⅱ+ low concentration Ginaton group. The concentrations of endothelin in culture liquid were measured by the method of radio-immunity. Results:①Ang Ⅱ induced significant secretion of endothlin by the vascular endothelial cells. ②Tea polyphenols and Ginaton had the effect to inhibit the VEC secreting ET by Ang-Ⅱ (P 〈 0. 01 ). ③Low concentration Tea polyphenols group and low concentration Ginaton group had more inhibiting effect compared with that of high concentration groups(P 〈 0. 01 ). Conclusion :①Ang II is capable of increasing secretion of endothlin by the VEC. ②Tea polyphenols and Ginaton can provide protection against high level of Ang II caused by simulated microgravity.
