水貂成骨细胞的原代培养及其鉴定

摘要 为探讨原代成年水貂成骨细胞的培养方法,建立细胞库。无菌条件下取7月龄成年水貂肋骨,剪碎,用胰酶、胶原酶消化后接种于培养皿内培养,获得原代细胞。通过细胞形态学观察及碱性磷酸酶染色、骨钙素免疫组化、钙结节染色进行鉴定。结果,所培养的细胞具有典型的成骨细胞形态特征,碱性磷酸酶染色、骨钙素染色、钙结节染色均呈阳性。证明培养出的细胞是成骨细胞。 To investigate primary osteoblast culture of mink rip,and establish a foundation for further experimental study.The costal bones from 7 months old minks were stripped off all soft tissues including the periosteum,rinsed and cut into small bone pieces under sterile condition.After digested by trypsin and collagenase,the pieces were put into the culture dishes.The acquired cells were identified based on the phenotype,ALP stain,BGP immunochemistry of osteoblasts and alizarin red stain of mineralization nodules.The results showed the cells obtained from mink costal bone were identified to be osteoblasts by inert microscope,ALP stain,immunochemistry stain of os teoblasts and alizarin red stain of mineralization nodules.These results indicated the cells obtained from mink costal bone having typical osteoblasts' characteristics.
