Taurolidine联合X-射线对前列腺癌细胞周期进程的影响

摘要 目的观察taurolidine联合X射线照射对前列腺癌(DU145)细胞周期进程的影响。方法通过流式细胞仪检测不同浓度的taurolidine单独或分别联合12、和4Gy的X-射线照射对DU145细胞周期的影响。结果50μmol/L的taurolidine可诱导DU145细胞发生G0/G1期阻滞,和s期的降低,并呈显著的时效和量效关系,当25μmol/L的taurolidine联合2和4Gy的X-射线照射时,细胞发生G2/M期阻滞,然而当50μmol/L的taurolidine联合2和4Gy的X-射线照射,细胞发生G2/M期阻滞的去除。结论taurolidine联合X射线照射导致前列腺癌细胞G2/M期阻滞的去除,可能是诱导其细胞凋亡的一个主要机制。 Objective To evaluate the effect of taurolidine in combination with X-irradiation on cell cycle progress of DU145 cells.Methods The redistribution of cell cycle induced by taurolidine alone or in combination wirh X-irradiation was evaluated in DU145 cells with flow cytometry assay.Results The taurolidine alone showed a dose-dependent arrest of G0/G1 phase in DU145 cells,whereas the arrest of G2/M phase was found by the combination of 25 μmol/L taurolidine with 2 and 4 Gy irradiation,but a evidengt decline of arrest in G2/M phase was induced by the treatment after the combination of 50 μmol/L taurolidine with 2 and 4 Gy irradiation.Conclusion Taurolidine in combination with irradiation induced the abrogation of G2/M phase arrest,which may be a main mechanism of its inducing apoptosis.
