大肠癌COX-2和PPARδ基因的表达及其与临床病理的关系

摘要 目的研究大肠癌组织环氧合酶2(COX-2)和过氧化物酶体增殖物激活受体δ(PPARδ)基因mRNA的表达水平及其与患者临床病理参数的关系。方法采用实时荧光定量道转录聚合酶链反应(RT-PCR)技术检测38例手术切除的大肠癌及相应癌外正常大肠组织中COX-2mRNA和PPARδ mRNA的表达。结果①表达阳性率COX-2mRNA在癌组织高于相应癌外正常组织(86.8%vs52.6%,P=0.001),PPARδ mRNA在两者间无差异(50.0%vs44.7%,P=0.646)。大肠癌组织COX-2mRNA的表达水平高于癌外正常组织[0.011(0,0.088)vs0.0016(0,0.017),P=0.036];PPARδ mRNA则两者间表达无差异[0.086(0,0.24)vs0.052(0,0.19),P=0.73]。②大肠癌浸润越深,COX-2mRNA相对表达水平越高,PPARδ mRNA的表达与临床病理参数间无明显关联;③COX-2和PPARδ mRNA表达无相关性(r=0.098,P=0.560)。结论大肠癌COX-2mRNA高表达,且与大肠癌浸润程度有关;PPARδ mRNA表达无明显上调,两基因间表达水平无相关性。 Objective To investigate the expressions of cyclooxygenase ( COX - 2 ) and peroxisome proliferator - activated receptor (PPARδ) mRNA in eolorectal cancer (CRC) and their correlations with clinieopathologic parameters. Methods Fresh tumor tissues and matched distal normal eoloreetal tissues were collected from 38 confirmed CRC patients undergoing surgical operation. The expression levels of COX -2 and PPARδ mRNA were quantified by using SYBR Green real - time RT - PCR. Results ①The positive rate of COX - 2 mRNA in CRC tissues was significantly higher than that in the corresponding normal colorectal tissues (86.8 % vs 52.6%, P = 0. 001 ). There were no differences in PPARδ mRNA positive rates between CRC tissues and normal tissues (50.0% vs 44.7% , P =0. 646). The COX -2 mRNA expressions upregulated significantly in CRC tissues [ 0.011 (0, 0. 088 ) ] , compared to those in the corresponding normal eoloreetal tissues [ 0. 0016 (0, 0.017 ) ] ( P = 0. 036). There were no significantly differences in PPARδ mRNA expressions between CRC tissues [ 0. 086 (0, 0.24 ) ] and normal tissues [ 0. 052 (0, 0.19 ) ] ( P =0. 73 ) ; ②The COX -2 RNA expressions were positively related with the depths of tumor invasion. However, the PPARδ mRNA expression levels were no correlations with elinieopathologie parameters; ③There was no correlation between COX -2 and PPARδ expressions. Conclusion High COX - 2 mRNA expression level is related with the invasive potential of CRC. PPARδ mRNA expression is not statistically different from that in normal mucosa. There is no correlation between COX -2 and PPARδ expressions.
