摘要
[目的]为环境监测提供灵敏的监测指标及指示植物。[方法]分别采用分光光度法和比色法测定了菹草叶片的叶绿素含量和可溶性蛋白含量,进而研究了Cu2+胁迫下菹草叶绿素及可溶性蛋白含量的变化规律。[结果]在整个试验过程中,对照组的菹草叶片始终呈翠绿色。Cu2+浓度为1.0 mg/L时,第2天菹草老叶褪绿,第3天菹草嫩叶开始褪绿,第4天菹草芽心褪绿。0.5 mg/L Cu2+处理8 d对菹草的影响不明显。在5种不同浓度的Cu2+胁迫下,菹草叶绿素a、叶绿素b、总叶绿素的含量均呈下降趋势且下降趋势随着处理时间的延长而愈加明显;菹草可溶性蛋白含量也呈下降趋势且其含量与胁迫时间和Cu2+浓度呈显著负相关关系。[结论]菹草能提供灵敏的监测指标,可作为环境监测的指示植物使用。
[ Objective ] The research aimed to provide sensitive monitoring index and indicator plant for environmental monitoring. [ Method ] The contents of chlorophyll and soluble protein in Potamogeton crispus leaves were measured by spectrophotometry and colorimetry resp. and then the variations of content of chlorophyll and soluble protein in P. crispus under Cu^2+ stress were researched. [ Result] In the whole experiment process, the P. crispus leaves in the control group were always emerald. When the conch, of Cu^2+ was 1.0 mg/L, the old P. crispus leaves became chlorotic on the 2nd day, its tender leaves began to become chlorotic on the 3rd day and its central buds became chlorotic on the 4th day. Treatment with 0.5 mg/L Cu^2+ for 8 d showed no obvious influence on P. crispus. Under the stress from Cu^2+ at 5 different concn. , all the contents of chlorophyll a and b and total chlorophyll in P. crispus showed decreasing trend and this trend was more and more obvious along with the extension of treating time ; its content of soluble protein also showed decreasing trend and the content showed negative relationship with stress time and Cu^2+ concn. [ Conclusion] P. crispus could provide sensitive monitoring index, so it could be used as indicator plant for environmental monitoring.
