摘要
[目的]探讨卵母细胞扫描电镜和透射电镜的制作方法,为相关研究提供技术支持。[方法]以猪GV期和体外成熟MII期卵母细胞为研究对象,分别制作扫描电镜样本和透射样本,并观察其超微结构变化。[结果]所用方法需样本量少,电镜下细胞形态结构完好,超微结构清晰。[结论]证实该方法适用于卵母细胞超微结构观察的制样。
[ Objective ] The research aimed to explore the manufacturing methods of scanning electron microscope (SEM) and transmission electron microscopy (TEM) for oocyte and provide technical support for related research. [ Method] With GV stage and MII stage oocyte as the study object, samples of SEM and TEM were prepared respectively, then changes of the ultrastructure were observed. [ Result ] The results showed that the method need small number samples, keep intact cell morphology and can see clear uhrastructure. [ Conclusion] The method is suitable for uhrastructural observation of oocyte.
