摘要 建立成年营养性肥胖SD大鼠模型,研究茶多酚对肝脏组织结构的影响。将60只21日龄SD大鼠,按体重相近原则分为普通对照组、模型对照组、试验Ⅰ组、试验Ⅱ组、试验Ⅲ组和试验Ⅳ组,试验期80d。普通对照组饲喂基础日粮,其它各组饲喂高能高脂日粮,40d后,试验Ⅰ、Ⅱ、Ⅲ、Ⅳ组分别灌服10、20、40、80mg/kg.d的茶多酚,普通对照组与模型对照组灌服等体积生理盐水,至80d试验结束后禁食不禁水12h,用10%水合氯醛麻醉(2.0mL/kg)。解剖,取每只SD大鼠肝脏适量,Bouin液中常温固定,梯度乙醇脱水,石蜡包埋,切片展片,HE染色,中性树胶封片,观察拍照。普通对照组的中央静脉形态清晰,肝细胞索结构完整,肝血窦形状正常;模型对照组的肝细胞固缩,肝细胞索狭窄,肝血窦增大,脂肪变性,大量炎性细胞浸润;试验组随着茶多酚剂量的增加,肝细胞体积逐渐增大,肝血窦逐渐减小,肝脏脂肪变性逐渐减轻,炎症现象逐渐减轻。茶多酚可增加肝组织中肝脂酶活性、降低肝组织中过氧化脂质含量,对脂肪肝有一定的防治作用。 According to the body weight,sixty rats,21-day-age SD,were divided into common control group,model control group,experimentalⅠ,Ⅱ,Ⅲ and Ⅳgroup to study the effect of tea polyphenols on the histo-structure of liver in nutritional obesity rats,and each group had 10 rats,while experimental period last 80 days.The rats in control group were fed in the basic ration,but the rats in model control group and experimental group were fed high energy-fat ration.After 40 days,the rats in experimental groupⅠ,Ⅱ,Ⅲ,Ⅳwere fed 10,20,40,80mg/kg·d tea polyphenols separately,the rats in commom control group and model control group were fed in the same volume of physiological saline.After 80 days,all rats,after they drunk water but ate nothing for 12 hours,were anesthetized with 10% chloral hydrate(2.0mL/kg),and dissected.Liver of each SD rat was collected and fixed in Bouin solution at room temperature,dehydrated with grads-alcohol,paraffin-embedded,stained by hematoxylin-eosin(HE),mounted with neutral rubber.The slices of liver was observed and pictured.Patterns of the central vein of rats in common control group were clear,the structural of liver cell cable was integrity,the shape of liver sinusoid was normal.Liver cells of rats in model control group were shrinked,the cord of liver cells was strictured,the liver sinusoids were increased,and a large number of inflammatory cells were infiltrated.With the dose of tea polyphenols increasing the volume of liver cell of rats in experimental group gradually increases,the liver sinusoid gradually decreases,the liver steatosis gradually reduce,and the phenomenon of inflammatory cells infiltration gradually reduces.The results showed that tea polyphenols can increase hepatic lipase activity and reduce lipid peroxide content of fatty liver.So tea polyphenols has a certain role in the fatty liver of prevention and treatment.
