摘要 目的Jagged1蛋白在大鼠肌源性干细胞(MDSCs)体外增殖分化为神经样细胞过程中的表达差异研究。方法大鼠乳鼠骨骼肌消化后体外培养增殖,第5代传代后细胞分为2组:对照组完全培养基培养,诱导组加入诱导剂诱导。6 d后观察2组细胞的形态变化,并进行NSE免疫细胞化学鉴定。然后通过免疫荧光观察Jagged1蛋白的表达差异。结果诱导组细胞NSE染色阳性,免疫荧光结果显示对照组Jagged1蛋白表达阳性,实验组基本不表达。结论大鼠肌源性干细胞分化为神经样细胞过程中Jagged1蛋白表达存在明显差异。 Objective To investigate the role of Jagged1 protein during the proliferation and differentiation of muscle derived stem cells (MDSCs) into neuronlike ceils in vitro. Methods The skeletal muscle stem ceils derived from the neonate rat were cultured and purified elonaily in vitro. The 5 th passage cells were divided into the control group and the experimental group. Morphological change and immunocytochemieal identification were observed six days after experiment intervention. was employed to examine the expression difference of Jagged1 protein. Results NSE positive cells were'detected in the experimental group. Immu- nofluorescenee showed that the levels of Jaggedl protein in the experimental group were much lower than that of the control group. Coll- ~lusions The levels of Jaggedl protein were decreased when the MDSCs differentiate into neuron - like ceils in vitro.
